Applications of the Quantitative B-hCG Hormone Measurement Kit in Serum or Plasma

This ELISA kit is used for confirming pregnancy, approximate gestational age determination, identifying ectopic pregnancy, assessing the likelihood of miscarriage, and monitoring trophoblastic tumors and hydatidiform mole. It is also used in evaluating response to treatment for conditions associated with abnormally increased or decreased hCG levels.

How to Use the Quantitative B-hCG Hormone Measurement Kit in Serum or Plasma

Allocate one well for the control, five wells for the standards, and wells for the number of samples. Place the remaining wells with desiccant inside the special bag and seal it. It’s best to test standards in duplicate.

Add 50 microliters of sample, standard, and control to the wells. Use a separate pipette tip for each. Add 50 microliters of assay buffer to all wells and gently shake the plate for 10 seconds. Cover the plate with a seal and incubate for 5 minutes at room temperature (20–25°C). Wash the wells 5 times with 300–350 microliters of wash solution (20–40 seconds each time). Then tap the plate on a paper towel to remove excess moisture. Add HRP conjugate: Add 100 microliters of conjugate to all wells and gently shake for 30 seconds. Re-incubate the plate for 5 minutes at room temperature. Repeat the washing step as in step 5. Add chromogen solution (TMB): Add 100 microliters of TMB solution to the wells. Incubate the plate for 5 minutes at room temperature, away from light, for the enzymatic reaction to produce a blue color. Add 50 microliters of stop solution to each well and mix gently. The blue color will change to yellow. Read the plate at a wavelength of 450 nm with a 630 nm reference. Do this within a maximum of 10 minutes after stopping the reaction.

Normal Values for the Quantitative B-hCG Hormone Measurement Kit in Serum or Plasma

In healthy non-pregnant individuals, serum hCG concentration is usually less than 5 mIU/mL. In a normal intrauterine pregnancy, hCG concentration is measurable approximately 1.5-1 week after conception and is about 1 mIU/mL. The concentration of this hormone doubles every 1.5-3 days during the first 6 weeks of pregnancy. In late first trimester, hCG concentration reaches its peak, approximately 100,000 mIU/mL. Then, from the early second trimester, its level slowly decreases. It’s worth noting that the concentration of this hormone is approximately double in twin or multiple pregnancies. In ectopic pregnancies, the concentration of this hormone is lower compared to normal intrauterine pregnancy. Samples with B-hCG concentrations in the range of 5-10 mIU/mL are considered suspicious for pregnancy, and repeat testing after 48 hours is recommended.
